Wide-field optically sectioning fluorescence microscopy with laser illumination

We describe an extremely simple method by which optically sectioned fluorescence images may be obtained withconventional microscopes using laser illumination. A onedimensionalgrid pattern is introduced into the illuminationsystem, together with a rotating ground glass diffuser. Thiscauses an image of the grid pattern to be projected into thespecimen. Images taken at three spatial positions of the gridare processed in a simple manner to provide opticallysectioned images of ¯uorescent specimens.
